Pharmacognostical characteristic, metabolite profiling, and angiotensin converting enzyme inhibitory activity evaluation of the methanol extract of Peperomia pellucida (L.) Kunth herbs

Background: Peperomia pellucida (L.) Kunth is a herb can be found in various shaded and damp habitats in tropical and subtropical climate. This plant is traditionally used as a medicine in Indonesia such as dizziness, headache, fever, gout, and hypertension. Objective: This study aimed to determine of pharmacognostic characteristic and angiotensin converting enzyme (ACE) inhibitory activity evaluation of P. pellucida methanol extract. Method: Extraction process was performed using gradual maceration (n-hexane and methanol). Pharmacognostic characteristic was determined using some parameters including metabolite profiling, total phenolic and flavonoid content, and physicochemical characteristic. Also, ACE inhibitory activity was evaluated using KIT ACE WST1 and microplate reader 96 wells. Results: In this work was obtained the yield of methanol extract (10.4%w/w). Metabolite profiling on methanol extract was showed a peak depth with a well-separated Rt (retention time) value ranging from 3.5 to 8.0 minutes which means that the extracted compound was from polar to nonpolar properties. Phenolic content and flavonoid content in methanol extract were 4,33% (as gallic acid) and 2,53% (as quercetin), respectively. Physicochemical properties were a specific odor, a distinctive taste, a greenish brown, the pH of 6.88, the water content of 3.62%v/w, the total ash content of 1.68%, the acid insoluble ash content of 0.05%, the ethanol soluble extractive of 82.46%, and the water soluble extractive of 16.74%. Whereas, ACE inhibitory activity (IC50) was 22.80 µg/ml. Conclusion: based on the results studies, the methanol extract had a potential as ACE inhibitory activity with the specific pharmacognostic characteristic.
