 Peperomia pellucida L. Kunth herb traditionally is used to decrease blood pressure since a long time ago. But, the scientific data as antihypertension is limited. As scientifically, based on the results of preliminary research, it is known that P. Pellucida L. Kunth herbs extract contains secondary metabolites that are rich in flavonoids and alkaloids constituents suspected of having contributed as an ACE inhibitor. The separation of alkaloids from natural resources is not easy and needed special methods to attract the maximum alkaloid. The aim of this study was to investigate the effect modification methods extraction of P. pellucida L. Kunth herbs for alkaloid enrichment and their TLC profiles. Dried P. pellucida L. Kunth herbs were extracted with several methods; method A (maceration using acid-base extraction with n-hexane and dichloromethane), method B (soxhlet using acid-base extraction with methanol and liquid – liquid extraction with dicloromethane), method C (maceration using acid-base extraction with dichloromethane), method D (reflux using acidic solution followed liquid – liquid extraction with dichloromethane) and method E (acid-base extraction using sulphuric acid followed by partition with dietyl ether). Each extract solution obtained was evaporated to obtained dry extract and analyzed the content of alkaloid and TLC profile. The results showing a difference from all modification of extraction method for alkaloids enriched, as well as TLC profiles.
